Gene targeting
Artificial modification of a gene in a specific and directed fashion. Typically refers to substituting one DNA sequence for another to inactivate a gene or introduce or correct a mutation in a gene.

New mutations at mouse loci can be generated by several techniques-radiation, chemical mutagenesis, or gene targeting.

Bone marrow stem cells from patients with this disease have had their mutant gene knocked out by gene targeting [Link] and gained the ability to make good collagen and bone (so far, only in mice).

Substitution of a segment of DNA by another that is identical (homologous) or nearly so. Occurs naturally during meiotic recombination; also used in the laboratory for gene targeting to modify the sequence of a gene. See recombination.
