Melting temperature At the melting temperature half of the oligo will form duplex with complementary oligo at the same concentration. PCR annealing temperature is usually set a few degrees lower than the melting temperature. See also PCR Glossary.
Melting temperature The temperature at which secondary or higher structures of a biological molecule are lost; for a nucleic acid, the melting temperature is defined as the temperature at which half the helical structure is lost.
The melting temperature of a primer is defined as the temperature at which 50% of that same DNA molecule species form a stable double helix and the other 50% have been separated to single strand molecules.
The choice of the length of the primers and their melting temperature depends on a number of considerations.
