Primer walking is a sequencing method of choice for sequencing DNA fragments between 1.3 and 7 kilobases.

 


Primer
A short oligonucleotide complementary to target DNA and acts as the leader for DNA extension. Also refers to short oligonucleotide used to primer reverse transcription of RNA to DNA in reverse transcription reactions. See PCR Glossary.

Definition of primer :
A short sequence (of RNA or DNA) from which DNA replication can initiate. May be either a synthetic DNA or RNA or a length of RNA synthesized in vivo by primase.

Primer
A primer is a short, single-stranded DNA sequence used in the polymerase chain reaction (PCR) technique. In the PCR method, a pair of primers is used to hybridize with the sample DNA and define the region of the DNA that will be amplified.

Primer Pheromones
Rats and mice give off pheromones that elicit mating behavior. However, the response is not immediate as it is in the releaser pheromones of mother rabbits and insects.

Short preexisting single-stranded polynucleotide chain to which new deoxyribonucleotides can be added by DNA polymerase. It anneals to a nucleic acid template and promotes copying of the template starting from the primer site.

Primers are short chains of about 20 nucleotides that are complimentary to a region in the DNA to be amplified. They are needed because the enzyme that copies the DNA (DNA polymerase) cannot start the process unless it has already been started.

In practice, when synthesising an oligonucleotide primer or probe based on this information a mixture of all the possible sequences is made, and referred to as a degenerate oligonucleotide.

As it cools, primers bind the DNA. These primers are short oligonucleotides chosen by the experimenter and added to the DNA mixture at the beginning. They flank the region to be amplified.

For the primers to anneal to the ssDNA fragments, the temperature is reduced to about 55° C. However, at this temperature the original complementary ssDNA fragments will begin to re-anneal with each other.

For instance replication requires a primer and specificity is determined by the sequence and size of the primer.
