Pernicious anemia is caused by a lack of intrinsic factor. Intrinsic factor is a protein produced by the stomach that binds to vitamin B12. The combination of vitamin B12 and intrinsic factor is absorbed in the lower part of the small intestine.

What is megaloblastic (pernicious) anemia?
Megaloblastic anemia, also called pernicious anemia, is a type of anemia characterized by very large red blood cells.

Absence of intrinsic factor, also called pernicious anemia - Intrinsic factor is a protein secreted by cells of the stomach lining. Intrinsic factor attaches to vitamin B12 and takes it to the intestines to be absorbed.

In elderly people, it is associated with pernicious anemia. Some people with this disorder have no symptoms, but many experience vague pain, a feeling of fullness, loss of appetite, belching, nausea and vomiting.

Vitamin B12 deficiency anemia (or pernicious [pur-NISH-us] anemia). This type of anemia happens due to a lack of vitamin B12 in the body. Your body needs vitamin B12 to make red blood cells and to keep your nervous system working normally.

Other less-common forms of anaemia include pernicious anaemia (inability of the body to properly absorb vitamin B-12), sickle-cell anaemia (an inherited disorder) and thalassemia anaemia (also inherited).

For example, an immune system disorder called pernicious anemia prevents normal absorption of vitamin B-12. Stomach or intestinal illness, some medicines, and some inherited problems may also cause vitamin B-12 deficiency.

People who have had stomach surgery or have pernicious anemia (severe decrease in red blood cells) or achlorhydria (absence of hydrochloric acid in the gastric juices, which help digest food) have an increased risk of stomach cancer.
